The lesions described relate principally to medical non-neoplastic conditions as exemplified by the first three Fascicles on endocrine, pulmonary, and skin diseases. Many of these lesions represent complex entities and when appropriate the authors have included contributions from internists, radiologists, and surgeons.

The earliest work in the field was primarily in the hands of botanists and many of the approaches to the under standing of the chemical composition of cells and tissues involved techniques such as microincineration, which destroyed structural integrity.
